RESUMO

Despite the advent of precision therapy for breast cancer (BRCA) treatment, some individuals are still unable to benefit from it and have poor survival prospects as a result of the disease's high heterogeneity. Cell senescence plays a crucial role in the tumorigenesis, progression, and immune regulation of cancer and has a major impact on the tumor microenvironment. To find new treatment strategies, we aimed to investigate the potential significance of cell senescence in BRCA prognosis and immunotherapy. We created a 9-gene senescence-related signature. We evaluated the predictive power and the role of signatures in the immune microenvironment and infiltration. In vitro tests were used to validate the expression and function of the distinctive critical gene ACTC1. Our risk signature allows BRCA patients to receive a Predictive Risk Signature (PRS), which may be used to further categorize a patient's response to immunotherapy. Compared to conventional clinicopathological characteristics, PRS showed strong predictive efficacy and precise survival prediction. Moreover, PRS subgroups were examined for altered pathways, mutational patterns, and possibly useful medicines. Our research offers suggestions for incorporating senescence-based molecular classification into risk assessment and ICI therapy decision-making.

Releasing Wolbachia-infected male mosquitoes to suppress wild female mosquitoes through cytoplasmic incompatibility has shown great promise in controlling and preventing mosquito-borne diseases. To make the release logistically and economically feasible, we propose a saturated release strategy, which is only implemented during the epidemic season of mosquito-borne diseases. Under this assumption, the model becomes a seasonally switching ordinary differential equation model. The seasonal switch brings rich dynamics, including the existence of a unique periodic solution or exactly two periodic solutions, which are proved by using the qualitative property of the Poincaré map. Sufficient conditions are also obtained for determining the stability of the periodic solutions.

In recent years, skutterudite filled with electronegative elements (S, Se, Cl, Br) has attracted the extensive attention of researchers. By doping electron donors (Pb, Ni, or Te, S, Se) at the Co or Sb sites, the electronegative elements can form thermodynamically stable compounds in the intrinsic pores of the skutterudite, substantially expanding the research scope of skutterudite. In this study, S0.05Co4Sb11.3Te0.6Se0.1 skutterudite was synthesized at high pressure and high temperature, with a pressure range of 2.0-3.5 GPa. The phase composition, micro-morphology, and electrical and thermal transport properties were systematically characterized. The micromorphology analysis shows that the introduction of S element and substituting Te and Se at the Sb sites inhibit the grain growth in a suitable high-pressure environment. Substantial differences are observed in the directions of the lattice stripes in the samples, and rich grain boundaries and many lattice distortions and dislocation defects occur. The carrier concentration can be optimized by filling the voids of the skutterudite with a few S atoms, and the thermoelectric properties can be optimized by scattering phonons through resonance scattering and defect scattering. The samples synthesized at a pressure of 3.0 GPa and a temperature of 900 K have a maximum power factor of 23.85 × 10-4 W m-1 K-2 and a maximum zT value of 1.30 at a test temperature of 773 K.

Nonexpressor of pathogenesis-related (NPR) genes are bona fide transcription cofactors in the signal transduction pathway of salicylic acid (SA) and play critical regulatory roles in plant immunity. However, the NPR family genes in Brassica juncea var. tumida have not yet been comprehensively identified and analyzed as of yet. In the present study, NPR genes in B. juncea var. tumida seedlings were identified, and the tissue-specific expression patterns of NPR genes in the seedling were analyzed under salt stress (200 mM) treatment and infection by Plasmodiophora brassicae. A total of 19 NPR family genes clustering into six separate groups were identified in the genome of B. juncea var. tumida. These BjuNPR family genes were located in 11 of 18 chromosomes of B. juncea var. tumida and each possessed 1-5 exons. The BjuNPR family members had similar protein structures and conserved motifs. The BjuNPR genes exhibited tissue-specific expression patterns in the root, stem, leaf, flower and pod. Some BjuNPR genes were sensitive to salt stress and showed up-regulated or down-regulated expression patterns and most BjuNPR genes were up-regulated upon infection by P. brassicae. This study provides a foundation for further research into BjuNPR genes regulation in plant growth, development, and abiotic stress tolerance.
